    Novel Vaccine for Chagas Disease: Efficacy Testing in Baboons, Descrição: Over the years, various groups have developed experimental vaccines, using mainly parasite lysates, or purified or recombinant proteins and peptides, and DNA, targeting almost exclusively protein antigens. Most, if not all, of these vaccines, however, have failed to provide sterile protection to animals challenged with different parasite strains. Although glycoconjugates (i.e., glycolipids and glycoproteins) are the major antigens on T. cruzi surface and contain highly immunogenic epitopes, they remain practically unexplored as vaccine targets. Patients with acute or chronic ChD, from diverse geographical locations in Latin America, have very high titers of trypanolytic anti--galactosyl antibodies (Ch anti--Gal Abs). These antibodies are protective and thought to be the major host immune mechanism controlling parasitemia at both acute and chronic stages of ChD. Ch anti--Gal Abs recognize highly immunogenic terminal, non-reducing -Gal-containing epitopes or glycotopes abundantly expressed on major immunodominant glycoproteins (i.e., mucins and TS/gp85 glycoproteins) of the mammal-dwelling trypomastigote forms. Since these -Gal epitopes are absent in human cells, we propose that an effective ChD vaccine could be obtained using these immunodominant B-cell epitopes. The experimental glycoconjugate or neoglycoprotein (NGP) vaccine, developed by Dr. Almeida (PI) and Dr. Katja Michael (Chemistry Dept., UTEP) (Co-PI), is based on a linear glycan (Gal1,3Gal1,4GlcNAc, or Gal3LN or KM24) that exists naturally on the surface of T. cruzi, and does not exist in humans and Old World primates such as baboons. Therefore, this T. cruzi sugar is detected as a foreign substance and a powerful immune response is mounted against it in patients with ChD and in transgenic mice lacking this glycotope. After experimental immunization with KM24 coupled to a carrier protein (bovine serum albumin, BSA) (NGP KM24b), the immune system of the immunized animals are primed to recognize T. cruzi as foreign and to kill the parasites soon after they enter the blood stream, and before they can get into cells where they reproduce. The KM24b vaccine also induces strong CD4 (helper) T cell activation, as observed with other glycoconjugate vaccines, and production of cytokines essential for isotype switching (from IgM to IgG) and differentiation of activated B cells into memory B cells, thus leading to a long-term immunity. Animals vaccinated with KM24b show 100% protection against two consecutive lethal parasite challenges and 99.9% parasite load reduction in the heart, as measured by qRT-PCR. Finally, lytic anti--Gal Abs purified from KM24b-immunized mice, are able to partially protect mice against T. cruzi infection, indicating a potential therapeutic value of this NGP vaccine. In addition to the KM24h vaccine, Dr. Almeida and collaborator (Dr. Rosa A. Maldonado, UTEP) have developed a peptide vaccine (MASPpep), based on a highly abundant and immunogenic surface antigen (mucin-associated surface glycoprotein, MASP), secreted by infective T. cruzi mammal-dwelling trypomastigote forms. MASPpep differs from previous peptides used as experimental ChD vaccines, because it is a longer (20-mer) peptide containing multiple B cell and CD4 and CD8 T-cell epitopes. Thus, it elicits a strong anti-T. cruzi immune response in mice mediated by both B and T cells, resulting in a very good protection (~86%) against lethal parasite challenge. Mice vaccinated with MASPpep also show 97% reduction of parasite load in the heart.     Synthetic neoglycopeptides as Chagas disease vaccines, Descrição: Chagas disease (ChD) is a devastating neglected tropical illness caused by the protozoan parasite Trypanosoma cruzi. It is estimated that 8-11 million people are chronically infected in Latin America, representing a substantial social and economic burden. Owing to the extensive global migration of asymptomatic, chronically infected individuals from endemic regions, ChD now affects thousands of people in nonendemic regions like North America and Europe. The two available drugs for ChD chemotherapy have limited efficacy in the chronic phase of the disease and are rather toxic. Moreover, there is no preventive or therapeutic vaccine for human ChD, despite numerous experimental efforts targeting almost entirely parasite proteins. Although glycoconjugates are the major surface molecules of the infective trypomastigote stages (from both insect vector and host cells), they remain completely unexplored as vaccine targets. Highly abundant T. cruzi trypomastigote surface glycoconjugates, such as glycosylphosphatidylinositol (GPI)-anchored mucin glycoproteins (tGPI-mucins), contain highly immunogenic terminal -galactopyranosyl residues (-Gal), which are not expressed in human cells. Accordingly, very high levels of trypanolytic, protective anti--Gal antibodies are observed in both acute and chronic stages of human ChD. We thus hypothesized that terminal -Gal-containing epitopes could be highly effective vaccines for ChD. To test our hypothesis, we recently employed the 1,3-galactosyltransferase-knockout (1,3GalT-KO) mouse model, which closely mimics the human humoral immune response against T. cruzi. As immunogen, we used the synthetic trisaccharide Gal1,3Gal1,4GlcNAc (Gal3LN), which is one of the major O-glycans on tGPI-mucins, covalently linked to bovine serum albumin (BSA) as carrier protein. Gal3LN-BSA-immunized mice showed very high levels of protective anti--Gal antibodies, much lower parasitemia, and very low parasite levels in the heart, as compared to control BSA-immunized animals. Although not sterile, the protection induced by the Gal3LN-BSA vaccine was long lasting and dependent on CD4+ (helper) T cells. Based on these results, we hypothesize that the ideal vaccine for ChD should induce both humoral (B cell) and cellular (T cell) immune responses, since both lytic antibodies and T cells play a decisive role in controlling T. cruzi infection. Aiming at the generation of a ChD vaccine that provides sterile protection against T. cruzi mediated by both B and T cells, here we plan to synthesize a series of neoglycopeptides (NGPs) consisting of a well-characterized major histocompatibility complex class II (MHCII)-binding peptide, i.e., tetanus toxoid peptide (TTp) or ovalbumin peptide (OVAp), covalently linked to the immunodominant Gal3LN epitope. These synthetic NGPs will be employed for the immunization of 1,3GalT-KO mice and the B and T cell-mediated immune responses will be examined. To achieve these objectives, we propose three specific aims: Specific Aim 1: Chemical synthesis of 1,3-Gal-containing neoglycopeptides. Specific Aim 2: Vaccination of 1,3GalT-KO mice with synthetic 1,3-Gal-containing neoglycopeptides and evaluation of the protective B cell-mediated immune response. Specific Aim 3: Elucidation of T cell-dependent immune mechanisms by synthetic T. cruzi glycan epitopes. We anticipate that high levels of protective anti--Gal antibodies will be achieved following immunization with the synthetic NGPs.     Global Characterization of Protein Palmitoylation in Trypanosomes, Descrição: Dual acylation with myristate and palmitate is essential for targeting of some proteins to specific lipid domains, and is commonly observed on eukaryotic cell signalling molecules. While N-myristoylation is carried out by a single N-myristoyltransferase, S-palmitoylation is mediated by one of a number of palmitoyl acyltransferases (PAT), each having its own subcellular location and substrate specificity. Palmitoylation is an essential modification in all eukaryotes. One goal of our research is to define for the first time the subcellular locations and substrates of every PAT in a eukaryotic cell. We have chosen the single-celled eukaryote Trypanosoma brucei for our studies, an excellent model system for this kind of analysis given its complex cell structure and powerful genetic tools. Over half a billion people are at risk of infection with one of three neglected tropical pathogens: Trypanosoma brucei, the agent of African sleeping sickness (60 million), Trypanosoma cruzi, the agent of Chagas disease (100 million), and Leishmania species (350 million), the agents of cutaneous, visceral and mucocutaneous leishmaniasis. There is no vaccine to prevent and/or treat any of these neglected diseases. Moreover, the drugs available for treating these infections suffer from poor efficacy and high toxicity. There are species-specific PATs in all organisms. Therefore, PAT inhibition may be a potentially novel drug target for the treatment of these and other infections with eukaryotic pathogens. A complementary aspect of our work will be the identification of those palmitoylating enzymes that are necessary for parasite growth and survival during infection and are therefore potential drug targets.
