Mechanisms of Arrhythmias Induced by Calcium-mediated Ectopic Beats in Post-infarction Hearts, Descrição: Cardiovascular diseases (CVDs) continue to be the leading cause of morbidity and sudden death in adults, exceeding the mortality associated with breast, lung, and colon-rectum cancers combined. Studies indicated that the majority of sudden cardiac deaths are attributed to ventricular tachycardia or fibrillation. The mechanisms governing the initiation and persistence of such cardiac arrhythmias are highly complex. They involve interactions across multiple scales ranging from genes to the whole organ. Therefore, there is a compelling need for novel technologies to bridge the gaps between scales and provide insights into underlying mechanisms and potential targets to treat CVDs. Over the past years, computer modeling and simulation of cardiac electrophysiology has emerged as a powerful platform for the investigation of arrhythmias. Computer models provide a framework for the unification of diverse experimental findings in different scales. The goal of this project is to use multi-scale computer models to investigate the origin of scar-related arrhythmias induced by calcium-mediated ectopic beats. To achieve this goal we make use of a state-of-the-art modeling framework to build high resolution computer models based on the advanced medical imaging techniques available at the German Heart Institute Berlin.. , Situação: Concluído; Natureza: Pesquisa. , Integrantes: Fernando Otaviano Campos - Integrante / Titus Kuehne - Coordenador.

Calcium-mediated Ectopic Excitations in Cardiac Tissue, Descrição: Cardiac arrhythmias are responsible for more than 300,000 deaths per year in the US alone. Despite extensive research over many decades the underlying mechanisms for these arrhythmias are not completely understood. A common mechanism, believed to underlie a variety of arrhythmias, is the presence of ectopic focal excitations in the heart. These ectopic foci disrupt the normal sinus rhythm, and can produce triggered excitations which can lead to reentry and/or wave fractionation in the heart. Remarkably, it is not understood what determines the timing, location and morphology of these focal excitations. Many experimental studies have shown that abnormal calcium (Ca) cycling, at the single cell level, plays an essential role in the formation of these focal excitations. However, the detailed mechanisms linking subcellular Ca and focal excitations at the tissue and whole heart level is not known. The underlying mechanisms for ectopic foci are difficult to understand because of the vast length and time scales involved. Therefore, novel approaches are needed to bridge the range of scales involved in the problem. In this project we propose to develop a multi-scale computational framework that can be used to describe the properties of Ca mediated ectopic foci. Our aim is to explore how abnormal Ca cycling at the subcellular level can summate over thousands of cells to form ectopic foci in tissue. Our computer models will shed light on the underlying mechanisms by bridging the gap between ion channels, cell electrophysiology, and tissue scale electrical activity.. , Situação: Concluído; Natureza: Pesquisa. , Integrantes: Fernando Otaviano Campos - Integrante / Gernot P - Coordenador / Yohannes Shiferaw - Integrante.

Cardiac Conduction in the Right Atrial Isthmus, Descrição: Severe atrial conduction disturbances, namely atrial flutter and atrial fibrillation are associated with aging. Research on atrial conduction comprising functional and structural aspects associated aging is therefore of particular interest. The lateral and posterior part of the lower right atrium is seen as critical substrate for the genesis of intermittent block of conduction. Recent studies on the tissue matrix in several regions of the atria suggest, that excitation spread at a sub-millimetre size scale may differ substantially from activation maps taken from clinical recordings at a coarser size scale. However, clinical mapping systems capable of resolving activation sequences at this size scale will not be available in the foreseeable future. The aim of this project is to study and compare mechanisms of macro- and micro-propagation in the atrial isthmus by means of a structure-related computer model with high spatio-temporal resolutions. Tuning of the virtual tissue model is made by electrophysiological experiments. The results should push forward the knowledge of microstructure related conduction disturbances. A profound analysis of the signature of microstructure on signals measured in the extracellular space will open new aspects of diagnostic tools of arrhythmia analysis for future minimal-invasive catheter techniques.. , Situação: Concluído; Natureza: Pesquisa. , Alunos envolvidos: Doutorado: (3) .
