  Patógenos e vetores como alvos biológicos: estudo de interação e mecanismos de ação de produtos naturais e sintéticos, Descrição: A biotecnologia tem sido cada vez mais reconhecida como uma fonte potencial de alternativas para o controle de doenças tropicais, zoonoses e controle de pragas agropecuárias. No entanto, esse potencial depende de um profundo conhecimento dos fatores biológicos que regem as interações parasito/vetor. Este projeto une equipes interdisciplinares para a investigação destes mecanismos em diferentes sistemas, de modo a identificar alvos para ações inovadoras e intervenções biotecnológicas. Os problemas de interesse envolvem o estudo da variação gênica e estruturação das populações naturais, o processo de adaptação dos organismos, relações evolutivas e sistemática. Os mecanismos defensivos utilizados pelas plantas incluem repelentes, fagoinibidores, hormônios e anti-hormônios que podem ser imensamente úteis para proteção de plantas agrícolas e controle de insetos vetores de doenças. Serão investigados, por exemplo, os efeitos de metabólitos secundários de plantas sobre o controle de Aedes aegypti, Rhodnius prolixus e Lutzomyia longipalpis, incluindo fatores de variação gênica e estruturação de células animais e vegetais, gerando dados que possam contribuir no manejo de vetores de arboviroses, doença de Chagas e leishmanioses. Pretendemos também aprofundar o conhecimento a respeito do controle do sistema neuroendócrino de insetos vetores sobre o desenvolvimento de parasitos em seus hospedeiros invertebrados, utilizando técnicas de manipulação neuroendócrina para investigar a organização ultra-estrutural do trato digestivo e o impacto das alterações induzidas sobre as diversas formas evolutivas de parasitas. Espera-se identificar estratégias que promovam uma interrupção direta sobre os mecanismos fisiológicos específicos necessários para que o inseto possa realizar com sucesso sua metamorfose, reprodução, diapausa e comportamento, através de uma interferência com seu sistema endócrino. Técnicas de proteômica, metabolômica e metagenômica serão utilizadas para investigará alterações metabólicas induzidas por parasitas em insetos hospedeiros que possam atuar em seu comportamento reprodutivo, incluindo glândulas responsáveis pela liberação de feromônios sexuais, avaliando potenciais usos no controle da população de vetores. Outras doenças tropicais como malária e esporotricose também serão objetos de estudo, onde se buscará identificar marcadores genéticos, epidemiológicos, hematológicos e imunológicos que permitam o desenvolvimento de novos testes diagnósticos, prognósticos e de discriminação de patógenos em infecções mistas de relevância clínica em medicina tropical.
