Cristina Vieira

Full Professor at the University Lyon 1, in Lyon, France, since 2010; Master Degree at the University of Lisbon, Portugal (1992); and doctorate at École Doctorale Évolution Écosystèmes Microbiologie Modélisation from Université Claude Bernarde Lyon I (1996); HDR (French post-doctoral degree allowing its holder to supervise PhD students) since 2003; Group Leader of the team "Transposable Elements, Evolution, Populations" since 2008; CNRS Delegation (2008-2009); Member of the Institut Universitaire de France - Junior (2009 to 2014). Our research consists in the study of the impact of transposable elements in the evolution of genomes and populations. The research themes are developed with combined approaches of bioinformatics and molecular biology.

    The genetic basis of adaptation to new environments: the Asian tiger mosquito case, Descrição: Aedes albopictus (the Asian tiger mosquito) is an invasive species. From its cradle in Asia it has colonized Europe, America and Africa during the last 30 years, mainly through intercontinental trade in used automobile tires where females lay their eggs. In France its range at first limited to the South East is rapidly expanding. The goal of our project is to understand if the success of the colonizing populations of A. albopictus is due to particular genetic characteristics that distinguishes them from the Asian populations.     Transposable elements and hybrids incompatibility, Descrição: genet_inc.png
	It has been postulated in the literature that TE may be some how implicated in hybrid incompatibility, or that they are associated with speciation events. At the intra species level, this was typically shown in the case of the hybrid digenesis in Drosophila, in which several different TEs may be involved. We are addressing this question with two different models. One consists in the analysis of hybrids from D. mojavensis and D. arizonae. This two species diverged less the 1 Mya and it is still possible to produce hybrids in the laboratory. We are analyzing the dynamics of TEs in the hybrids versus the parental lines. The other deals with a lower level of divergence using intra-specific hybrids between distinct natural populations of D. melanogaster or D. simulans.     Transposable element dynamics and variability between and within species, Descrição: Transposable elements are found in the genomes of virtually all living organisms, but their proportions and activities vary greatly between and within species. In order to understand TE dynamics, we focus on wild-type strains of Drosophila melanogaster and D. simulans, two closely related species which have a three-fold difference in their TE content. In particular, we explore variability in epigenetic TE regulation in relationship to variability in TE dynamics and content. One of the major defense mechanisms against TEs in Drosophila is an RNA interference machinery based on a particular class of small RNAs: piwi-interacting small RNAs (piRNAs). We are currently investigating the link between differences in TE and piRNA content between different populations and species of Drosophila using available genomes and NGS data. We focus on tirant, an endogenous retrovirus which activity varies between wild-type strains of D.simulans: while tirant is either inactive and embedded in heterochromatin in most strains, or it displays post-transcriptionaly silenced euchromatic copies, we identified one strain in which tirant behaves as an endogenous retrovirus and increases in copy number.     Análise da dinâmica de aquisição e perda de elementos transponíveis no genoma: o elemento Helena de Drosophila como modelo, Descrição: O elemento transponível Helena propicia uma oportunidade única para compreender a dinâmica de aquisição e perda de elementos em um genoma, já que em D. melanogaster há uma provável inativação desse retroposon e em D. simulans, uma atividade potencial do elemento. Além disso Drosophila é um excelente modelo de estudo, pois a quantidade de elementos é suficientemente pequena para permitir estudos exaustivos e completos. Para compreender o processo dinâmico de aquisição e perda de Helena nos genomas, este trabalho tem os objetivos de analisar a ocorrência do elemento Helena, in silico e em populações naturais, de 12 espécies de Drosophila (D. melanogaster, D. simulans, D. yakuba, D. erecta, D. sechellia, D. ananassae, D. persimilis, D. pseudoobscura, D. virilis, D. mojavensis, D. grimshawi, D. willistoni). As análises de Bionformática permitirão descrever o número de inserções de Helena no genomas, a diversidade nucleotidica, as relações evolutivas. e: o grau de deterioração entre cópias da mesma espécie e entre espécies. As análises populacionais de D. simulans e de D. melanogaster, como também de espécies subgrupo melanogaster e de D. virilis por meio de Southern blot permitirá detectar possíveis cópias completas e analisar sua expressão por meio de RT-PCR e Northern blot. Com esse estudo, sobretudo pelas análises populacionais em espécies irmãs (D. melanogaster e D. simulans), mas que apresentam dinâmica diferente de evolução do retroposon Helena, poderemos compreender como as modificações da estrutura e a composição desse elemento, um fenômeno que ocorre em etapas e velocidades variáveis em diferentes espécies. Com este tipo de análise comparativa poderemos compreender como o elemento Helena, em particular, e os retroposons em geral, são adquiridos e perdidos pelas espécies e como características, tais como os sítios de inserção, o genoma hospedeiro e o tempo de integração das cópias completas no genoma hospedeiro relacionam-se com a sua dinâmica evolutiva.. , Situação: Concluído; Natureza: Pesquisa. , Integrantes: Cristina Vieira - Integrante / GRANZOTTO, ADRIANA - Integrante / CARARETO, CLAUDIA M.
