    Investigation of giant aggregates of beta-amyloid, Descrição: Stability of honeycomb beta-amyloid aggregates investigated by light microscopy and light scattering in comparison with general Mie scattering theory. In aqueous medium, the beta-amyloid peptide (Abeta) partly exists as large (ca. 100 micro-m) aggregates, which, when examined under light microscopy, resemble a honeycomb structure with repetitive elements of about 3 micro-m. In treatments of neuronal cultures with Abeta, these structures exhibit a remarkable longevity. Direct microscopic observation reveals a significant stability even in the presence of denaturating agents such as urea and guanidine hydrochloride. Right-angle light scattering experiments in the presence of these chaotropic agents seem to corroborate these results, if the changes in refractive index of the medium in the presence of denaturants are taken into account.     Fluorescence microscopy image analysis, Descrição: A routine for histogram analysis of images has been written in the object-oriented, graphical development environment LabVIEW. The program converts an RGB bitmap image into an intensity-linear grayscale image according to selected conversion coefficients. This grayscale image is subsequently analyzed by plots of its intensity histogram (the number of pixels plotted against their respective brightness levels) and probability distribution of the brightness, and by calculation of various parameters, including average brightness, standard deviation, variance, minimal and maximal brightness, mode, skewness and kurtosis of the histogram and the median of the probability distribution. The program allows the selection of a region of interest (ROI) in the image and/or lower and upper threshold levels for analysis (e.g., to permit the removal of a constant background signal). The combination of these analysis features in a single program allows fast analysis of relatively large sets of image data. The results of the analysis of multiple images can be saved for data transfer and plotting in other programs.
