Protozoário virulência: Meu laboratório centra-se na patogenicidade dos micróbios eucarióticos pesquisando Doença de Chagas, leishmaniose, doença do sono, giardíase, criptosporidiose, toxoplasmose e tricomoníase. Estamos interessados na biologia celular e molecular dos próprios micróbios e das respostas de acolhimento que eles provocam e nós procuramos identificar e explorar fatores de virulência para novas intervenções e marcadores como epidemiológicos e taxonômicos como ferramentas para informar a política de saúde pública.

Exploiting the host response to cell invasion by trypanosomes, Descrição: Interaction of Trypanosoma cruzi and host target cell may begin even prior to attachment, with secreted parasite products of the oligopeptidase B and parasite derived eicosanoid thromboxane A2 able to engender calcium mobilization. In addition, host modified substrates of the cysteine protease cruzipain, such as bradykinin, are able to initiate responses from its heterotrimeric G protein coupled receptor (GPCR). The trypanosome surface is an adherent glycocalyx, rich in glycophosphatidyl inositol (GPI)-linked glycoproteins and presumably binding firmly to host cell plasma membranes by cross-linking of glycans and lectins forming a tight synaptic junction the parasite synapse. At the parasite synapse, trypansome mucins, GIPLs (glycosinositol phospholipids), MASPs (mucin-associated surface proteins), DGFs (dispersed gene family), TS (trans-sialidase) and TSL (trans-sialidase-like) proteins display glycans which bind host lectins and oligomerize with host integrins and other GPI-linked glycoproteins of the plasma membrane. Sialic acid present on host glycans (but not synthesized by trypanosomes) can also be bound and transferred within the parasite synapse by the action of the TS, while TS, TSL and DGF proteins in particular appear to be lectins in their own right and are able to bind and further cross-link host glycans on GPI-linked glycoproteins and glycosphingolipids.T. cruzi mucins may also bind laminin via polyvalent galectin-3 oligomers present on the host cell surface or free in the extracellular milieu thus promoting parasite-cell adhesion. A resulting lattice of this type, formed on the outer leaflet of the plasma membrane, may consequently be regarded as a type of lipid raft. Trichomonas gallinae virulence and host susceptibility, Descrição: Avian trichomonosis, caused by the flagellated protozoan Trichomonas gallinae, is a recently emerged infectious disease of British passerines. The aetiological agent, a clonal epidemic strain of the parasite, has caused unprecedented finch mortality and population-level declines in Britain and has since spread to continental Europe. It is now represents a threat to columbids, raptors and passerine species worldwide. Detection and risk assessment of waterborne protozoan parasites, Descrição: The majority of the world s population still live without access to healthy water and the contamination of drinking water with protozoan pathogens poses a serious threat to millions of people in the developing world and the deaths of many hundreds of thousands of children each year. Even in the developed world periodic outbreaks of diarrhoeal diseases are caused by ingesttion of the chlorine resistant cysts protozoan parasites Cryptosporidium sp., Giardia duodenalis. Thus, surveillance of drinking water is imperative to minimize such contaminations and ensure continuous supplies of healthy water world-wide. Work in my laboratory is focussed on the molecular definition of virulence factors of Cryptospodium and Giardia. We use genomics and proteomics to identify these and aim to exploit them once validated for use in the definition of virulent strains, as targets for therapy and prophylaxis, for risk assessment and in the design or refinement of public health intervention or for use in monitoring water supplies. Kinetoplastid Cell Biology, Descrição: Kinetoplastids are evolutionarily divergent flagellates with a variety of single copy organelles that are replicated with fidelity once per cell cycle. They posess an elaborate mitochondrion which is regulated during the complex life-cycle during which extreme changes in morphology and biochemistry are co-ordinately regulated witht he life-cycle. Some kinetoplastids are able to adopt an intracellular life-cycle multiplying within the cells of the vertebrate host.
