Lack of brain serotonin affects feeding and differentiation of newborn cells in the adult hypothalamus, Descrição: Given the potential role of serotonin in energy balance (e.g. satiety), and altered energy expenditure in Tph2-/- mice, a direct approach is eminent. Here, we will investigate changes in food intake, and precursor cell proliferation and survival in the adult hypothalamus at baseline and following high cholesterol diet (HCD).. , Situação: Concluído; Natureza: Pesquisa. , Alunos envolvidos: Mestrado acadêmico: (1) . , Integrantes: Friederike Klempin - Coordenador / Marike van Lingen - Integrante., Financiador(es): Berlin Institute of Health - Auxílio financeiro.

Identifying the mechanisms of antidepressant drug action in mice lacking brain serotonin, Descrição: Major depression is a serious mental disorder, which affects up to 15% of the population. Pharmacological manipulation of serotonin (5-HT) leads to clinical improvement associated with a delayed increase in hippocampal neurogenesis. ?State of the art? therapy consists of selective serotonin reuptake inhibitors (SSRIs), however, not all patients respond to this treatment. Intriguingly, tianeptine, which acts as a selective serotonin reuptake enhancer (SSRE) is also a potent antidepressant and has therefore been called a ?hypothesis killer?. Here we propose that the lack of a clear mechanism for SSRI function significantly limits our ability to effectively manage depression. In this proposal we a) take advantage of our recent insights into the role of serotonin in activity-dependent neurogenesis, b) apply a powerful genetic model that lacks brain serotonin to test a variety of pro-, and antidepressive stimuli that will directly translate to the clinical situation, and c) explore the interplay of brain-derived neurotrophic factor (BDNF) and serotonin.. , Situação: Concluído; Natureza: Pesquisa. , Alunos envolvidos: Doutorado: (1) . , Integrantes: Friederike Klempin - Coordenador / Golo Kronenberg - Integrante / Markus Petermann - Integrante., Financiador(es): Berlin Institute of Health - Auxílio financeiro.

Serotonergic mechanisms and therapeutic relevance of neuron (re)generation in the adult brain, Descrição: Today?s serotonin pharmacotherapy is based on treatment outcome that lacks a clear understanding of the underlying therapeutic mechanisms but also the pathways that produce counter-indications. Here, I propose new insights will come from pharmacologically targeting receptors in combination with a powerful imaging approach for the in vivo assessment of mechanism of serotonin action.. , Situação: Concluído; Natureza: Pesquisa. , Integrantes: Friederike Klempin - Coordenador., Financiador(es): Deutsche Forschungsgemeinschaft - Auxílio financeiro.
