Skills: in vivo studies using mice models (handling soft tissues, mainly liver and kidney/ hard tissues, strictly dental samples). In vitro experiments such as cell culture (mesenchymal cells, fibroblasts, human breast adenocarcinoma cell lines, colon adenocarcinoma cell lines). Molecular analysis: Trypan blue based cell viability assay, ELISA kits, immunohistochemistry, immunoprecipitation, quantitative and qualitative PCR methods, histological analysis, flow cytometry, protein isolation, western blotting, zymography. Mass spec sample preparation and interpretation (Nano-LC-ESI-MS/MS), Proteomic analysis: gel electrophoresis, protein interaction networks, bioinformatic tools applications, (Cytoscape), Atomic force microscopy (AFM, nanoscope, Gwyddion).

Proteomic Analysis of Liver in mice with different susceptibilities to Fluorosis, Descrição: Concern over the possible carcinogenic potential of fluoride has been voiced over the past 25 years. It has also been suggested that there may be an association between lifetime exposure to low level fluoride in drinking water and the susceptibility to hip joint fracture in old age. Fluoride in various chemical forms, doses, and exposures has physicochemical and biologic effects on cells and tissues. A narrow therapeutic/toxicity window and biphasic actions further complicate our understanding of fluoride?s effects. Fluorides mediate their actions through MAPK signaling pathways, leading to changes in gene expression, cell stress, and even cell death. The uncertainty associated with increased intake of fluoride is a major area of concern in an aging world population and needs further clarification by detailed examination to reach a more definite conclusion. The wide spread use of F has added to caries decline more over the excessive intake may effect the bone metabolism and enamel development which ultimately leads to skeletal and dental fluorosis.Many sources are there for flouride intake such as drinking water,use of dental products, food, toothpastes and infant formulas [1].Strong evidences are there to prove that increase in prevalence of dental fluorosis is linked to both flouridated and non fluoridated communities [2]. Although the exact mechanism by which flouride affects bio mineralization process are still to be interpreted [3,4].It`s been already proposed that genetic determinants influence the susceptibility to dental fluorosis in humans [5] and mice [6] respectively.In this context of DF two strains of mice have been identified with different responses to the effects of F in the mineralized tissues.The A/J strain is "Susceptible" with the rapid onset of development of DF, while the 129P3/J is "resistant" with minimum development of DF [6] not only this the mentioned to strains also differ towards their susceptibilities to the effect of F in bone [7,8]. Evidences are also there to suggest that drinking water fluoride levels over 2.0mg/L can cause damage to liver and kidney functions in children and that the dental fluorosis was independent of damage to the liver but not the kidney [9].
