Sotiris Missailidis

Possui graduação em Biologia pela The American University of Athens (1991), mestrado em Química pela University of York (1993) e doutorado em Química pela University of York (1997). Pós-doutorado em Cambridge e Nottingham nas áreas de Ciências Farmacêuticas e Biologia Molecular, respectivamente, e experiência anterior como professor na The Open University do RU e professor visitante das Universidades de Lisboa, Paris e Patras, bem como fundador e Diretor da Euzoia Limited no RU. Premiado pela Sociedade Europeia Contra Cancer (EACR) chegou no Brasil em 2011 como pesquisador visitante do Universidade Federal do Rio de Janeiro e subsequentemente atuou como Pesquisador Visitante Sênior no Laboratório de Hantaviroses e Rickettsioses da FIOCRUZ, e Professor Visitante Sênior do Instituto de Biofísica Carlos Chagas Filho do Universidade Federal do Rio de Janeiro. Atualmente é Especialista e Vice-Diretor de Desenvolvimento Tecnológico de Bio-Manguinhos na FIOCRUZ. Tem experiência na área de Bioquímica, Biofísica e Química Bio-inorgânica, com ênfase em Biologia Molecular, atuando principalmente nos seguintes temas: aptâmeros, G-quadruplexes, inflamação e câncer e desenvolvimento de novos diagnósticos e terapêuticos a base de ácidos nucleicos e vacinas sintéticas.

Aptâmeros com bases de DNA modificadas para inativação da enzima heparanase como uma nova terapêutica do câncer, Descrição: No passado, o grupo desenvolveu vários métodos de preparação de oligômeros substituídos com várias bases modificadas, incluindo bases sensíveis às radiações de 340nm como a 4-tio- timina, 4-tio-5-bromo-timina e 6-tio-guanina e as utilizou para vários estudos bioquímicos. Por exemplo, oligômeros contendo 6-tio-guanina tem sido usado para promover a ligação cruzada do elemento de resposta sérica c-fos e para produzir danos sítio específicos na proteína PCNA que pertence à maquinaria de replicação de DNA humano. Oligômeros contendo 4-tio-timina, irradiados com fótons 340nm, também foram usados para provocar a morte de células de pele em cultura e oferecer um protocolo útil para atingir o vírus de papiloma humano (HPV). Baseado no trabalho anterior do grupo da OPEN, propõe-se a modificação dos aptâmeros selecionados contra a enzima heparanase com tiobases inseridas em várias posições estratégicas dentro do aptâmero. Uma vez que os aptâmeros estejam desenhados e sintetizados pretende-se examinar sua propriedades físicas e químicas e suas interações com a proteína alvo utilizando uma série de técnicas. Uma vez definido o número e posições ideais de tio-bases no aptâmero, esta molécula modificada ligada à proteína alvo será exposta à radiação de 340nm e o efeito na estrutura da proteína alvo será avaliado. Com a exposição do tio-aptâmero à 340nm, ele será ativado devido a absorção de energia da radiação e consequentemente poderá ser reagir com outras moléculas como o O2, produzindo radicais os quais pode produzir danos à moléculas vizinhas.
