Mechanisms mediating anti inflammatory effects of omega 3 fattyacids in metabolic disorders: Role of lipid mediators and microRNAs (Sprint 3/2015), Descrição: Obesidade é uma doença complexa que aumenta a mortalidade e co-morbidades tais como diabetes e doenças cardiovasculares e caracteriza-se por expansão e inflamação do tecido adiposo. Estudos recentes mostram que mediadores lipídicos (leukotrieno B4 e Fator Ativador de Plaquetas, PAF) contribuem para a inflamação crônica que leva a obesidade e a resistência a insulina. Ácidos graxos poli-insaturados de cadeia longa omega-3 (PUFA) derivados de óleo de peixe são potentes nutrientes anti-inflamatórios conhecidos por reduzir os níveis de prostaglandina E2, LTB4 e outros marcadores de inflamação. Interessantemente, o LTB4 controla o perfil de expressão de miRNAs e modula a resposta inflamatória de células da imunidade inata como macrófagos. Inflamação no tecido adiposo é mediada primariamente por macrófagos que infiltram no tecido adiposo. Entretanto, o papel de omega-3 em regular os mediadores lipídicos para prevenir ou tratar a inflamação crônica não e conhecido até o momento. Nossa hipótese é que o óleo de peixe reduziria a inflamação crônica associada com disfunções metabólicas diminuindo a adiposidade e a resistência a insulina e que estes efeitos seriam mediados por mecanismos que envolvam mediadores lipídicos como LTB4 e PAF e miRNAs reguladores. ASSOCIAÇÃO DE PRRs COM RECEPTORES PARA MEDIADORES LIPÍDICOS EM MACRÓFAGOS E CÉLULAS DENDRÍTICAS., Descrição: Macrófagos e células dendríticas apresentam receptores de membrana(PRRs) que reconhecem estruturas microbianas, a família de receptores toll-like sendo os receptores prototípicos deste grupo. Outro grupo de PRRs reconhece moléculas e células próprias alteradas tais como lipídeos oxidados e células mortas e, deste grupo, o CD36 é o receptor mais estudado. A estimulação destes receptores induz a síntese imediata de mediadores lipídicos (prostaglandinas,leucotrienos e PAF) seguido de transcrição de vários genes. Macrophages e DCs desempenham um papel central no desenvolvimento, homeostasia, reparo tecidual e resposta imune. Estas células possuem alta plasticidade mudando seu estado funcional de acordo com estímulos do microambiente. Evidências de nossos trabalhos e de outros sugerem que a estimulação autócrina de receptors para mediadores lipídicos, em associação com a ativação de PRRs, é capaz de modular o fenótipo de macrófagos e DCs. No presente estudo nos propomos a investigar o efeito da associação: a) PAFR/CD36 nas células do microambiente tumoral; b) PAFR/CD36 na imunossupressão associada a tumores; c) PAFR/CD36 na resposta de tumores a quimioterapia e irradiação; d) PAFR/TLR4 em células dendríticas e na resposta imune in vivo; e) PAFR e receptor para LTB4 com os vários TLRs no fenótipo de macrófagos; f) receptor para LTB4/TLR4 em macrófagos e seu efeito na inflamação e resistência a infecção em diabéticos. ASPECTOS MOLECULARES ENVOLVIDOS NA ATIVIDADE MICROBICIDA E INFLAMATÓRIA DE LEUCÓCITOS NO PULMÃO, Descrição: O reconhecimento dos microorganismos pelos fagócitos envolve complexos de receptores(PRRs) que reconhecem padrões moleculares associados a patógenos (PAMPs), receptores para opsoninas (Fcg e C3b), receptores do tipo toll, além de receptores para mediadores inflamatórios. Existem evidências de interações entre os receptores da imunidade inata, receptores Fcg e receptores para eicosanóides, e que estas interações influenciam as funções efetoras dos macrófagos, tais como fagocitose e atividade microbicida. Estas interações podem ocorrer entre os receptores presentes em rafts lipídicos e/ou proteínas G envolvidas que levam a indução de diferentes vias de transdução de sinal elicitadas pela ativação destes receptores. Recentemente, demonstramos que o leucotrieno B4 aumenta a atividade microbicida de macrófagos alveolares infectados com Klebsiella pneumoniae (opsonizada com anticorpos) e que este aumento é dependente da ativação de PKC - delta e subseqüente ativação da NADPH oxidase. Não se sabe, entretanto se os leucotrienos interferem com a formação do fagolisossomo, um passo fundamental no controle de infecção por macrófagos. Assim, a primeira parte deste projeto tem por objetivo caracterizar as vias de sinalização acionadas em macrófagos alveolares pela ativação dos receptores Fcg, receptores tipo toll e receptores para leucotrienos, individualmente ou em associação, e as consequências destas associações na fagocitose, formação do fagossomo e atividade microbicida. Na inflamação pulmonar os leucócitos que migram para o pulmão são fundamentais na indução e resolução do processo inflamatório mas podem também ser importantes efetores de lesão tecidual. Trata-se de um título honorífico em virtude de minha atuação passada e presente no Depto de Farmacologia da Faculdade de Medicina da U. de Sherbrooke. Com o prof. Pierre Sirois daquele departamento iniciamos em 1989 um Convênio bilateral CNPq/MRC (Canadá) que vigorou por mais de 10 anos. Durante minha estadia no departamento que foi de 15 a 30 dias por ano, ministrei aulas de pós graduação, co-orientei mestrandos e doutorandos daquele departamento, O Pierre Sirois co-orientou alunos meus que fizeram estágios sandwich ou pós-doutorado.
