Bioprospecting hydroxylated chalcones in in vitro model of ischemia-reoxygenation, Descrição: Ischemia reperfusion (I/R) is the main cause of acute kidney injury, contributing to higher rate of mortality and morbidity around the globe. Renal I/R injury involves complex pathophysiology. However, generation of reactive oxygen species (ROS) are supposed as the common pathological alteration during I/R injury. Therefore, targeting generation of ROS, protecting mitochondria and improving organ function are the main interventions to manage I/R induced AKI. Chalcones belonging to flavonoid family of phenolic compounds are abundantly found in numerous fruits, dietary spices, teas and soya-based foods. Chalcones derivatives are studied widely due to their simple structure and numerous biological activities such as antioxidant, anti-inflammatory, anti-microbial, anticancer, antidiabetic, psychoactive and neuroprotective actions. Therefore, considering strong antioxidant action along with other pharmacological roles of chalcone derivatives, 2-hydroxy chalcones were synthesized and then analyzed for their nephroprotective role in I/R induced AKI. The main objective of the study was to evaluate the role 2-hydroxy chalcones in treatment and prevention of I/R induced AKI by targeting main pathological pathways including generation of ROS, apoptosis, necrosis and antioxidant system. Cell viability was analyzed via MTT assay, followed by flow cytometry for apoptosis, necrosis, ROS generation and mitochondrial transmembrane potential measurement. The affect of chalcones on oxidation reduction system was also evaluated by analyzing its effect on TBARS, GSH and SOD. The effect of chalcone on cells morphological features were studied via scanning electron microscopy. Nephroprotective role of Helianthus Annuus, Descrição: Helianthus Annuus seeds was collected and one part was extracted with methanol while the other part was extracted with n-hexane. Both of the extracts were applied in mice model for their nephroprotective role against gentamicin induced nephrotoxicity. The nephroprotecttive role was estimated from measuring blood biomarker (serum creatinine, BUN) and kidney histopathology.
