  Panorama imunológico, transcricional e mutacional do microambiente tumoral em carcinoma orofaríngeo de células escamosas associado ou não a infecção pelo HPV, Descrição: Os tumores de cabeça e pescoço ainda exibem uma alta incidência no Brasil e no mundo. O carcinoma orofaríngeo de células escamosas possui, como um dos seus fatores de risco, a infecção pelo vírus do papiloma humano (HPV). Pacientes com carcinoma de orofaringe associados a infecção pelo HPV geralmente apresentam um melhor prognóstico e probabilidade de sobrevida quando comparados aos pacientes não infectados pelo HPV. A resposta imune do hospedeiro, o panorama transcricional e mutacional podem esclarecer algumas dessas diferenças observadas entre os tumores de orofaringe relacionados ou não à infecção pelo HPV.   MISTIC-fusion proteins as antigens for high quality membrane protein antibodies, Descrição: Lack of high-quality antibodies against transmembrane proteins is a widely recognized hindrance in biomedical and cell biological research. Here we present a robust pipeline for the generation of polyclonal antibodies employing full-length membrane proteins as immunogens to overcome this "antibody bottleneck". We express transmembrane proteins fused to a MISTIC fragment that enhances expression of eukaryotic membrane proteins in E. coli. Purified membrane proteins are used as immunogen for rabbit injection employing standard immunizing protocols.   Crystal Structure of the Herpesvirus Nuclear Egress Complex Provides Insights into Inner Nuclear Membrane Remodeling, Descrição: Although nucleo-cytoplasmic transport is typically mediated through nuclear pore complexes, herpesvirus capsids exit the nucleus via a unique vesicular pathway. Together, the conserved herpesvirus proteins pUL31 and pUL34 form the heterodimeric nuclear egress complex (NEC), which, in turn, mediates the formation of tight-fitting membrane vesicles around capsids at the inner nuclear membrane. Here, we present the crystal structure of the pseudorabies virus NEC. The structure revealed that a zinc finger motif in pUL31 and an extensive interaction network between the two proteins stabilize the complex. Comprehensive mutational analyses, characterized both in situ and in vitro, indicated that the interaction network is not redundant but rather complementary.
