  Implications of RNA-seq in the detection of anabolic steroid use and harnessing of the molecular mechanism of muscle memory, Descrição: The variable effects of anabolic androgenic steroids (AAS) on increasing skeletal muscle mass and strength has been well documented, as is the misuse of AAS in sport. Recent evidence suggest either a long- or short-term exposure to AAS might have a sustained effect on muscle morphological changes, for example, with increased muscle mass, capillary per fibre, muscle fibre size and myonuclei density, leading to improved performance. A positive correlation between the number of myonuclei and training response following exposure to AAS in a mouse model seems to suggest a link between the formation of extra myonuclei and the extent of ?muscle memory?; an idea that requires further investigation in humans. Strength training can also increase the number of nuclei in muscle fibres. Adaptations in muscle mass by strength training are significantly enhanced in previously trained individuals despite a prolonged detraining period. Given the persistence of muscle nuclei, the use of AAS combined with training will have a greater impact on muscle hypertrophy than either training or steroid use alone. To detect the long-term effect of AAS and training (even after drugs are no longer detectable in the human system), abnormal changes in skeletal muscle morphology illustrated by specific gene markers in response to the stimuli exist and will allow these molecular signals to be picked up by modern gene screening methods. In the proposed project, gene expression profiling of skeletal muscle in response to AAS exposure will be carried out using total RNA-seq.   A systems biology biomarkers approach to the differentiation of recombinant human erythropoietin doping from confounding factors, Descrição: The current research on the molecular signature of rHuEPO doping has, so far, provided some evidence that ?omics? technologies such as transcriptomics have the potential to significantly strengthen the current ABP approach and contribute to other traditional anti-doping tests. This approach if successful can in the future be applied to the detection of other doping substances and methods difficult to detect such a recombinant human growth hormone and blood transfusions. There is also the interesting possibility that an "omics"-based approach could help reduce the pressure on the anti-doping obligations of athletes such as the ?athletes whereabouts?. In order to confirm that an integrative ?omics? approach is a possible solution to improve rHuEPO detection, it is of paramount importance to precisely determine normal gene expression reference values as well as to carefully assess the potential effects of external factors on blood gene expression profiles, such as prior training, altitude including different hypoxic ?dose? and protocols, sport discipline, level of competition, gender, ethnicity and age.   Projeto Atletas do Futuro - a Ômica no Esporte, Descrição: Após o término do Projeto Genoma Humano e o estabelecimento da sequência completa de nosso código genético, o interesse dos biologistas tem se deslocado para a análise de variações nas sequências não-repetitivas presentes no genoma. Os polimorfismos de nucleotídeos únicos (do inglês single nucleotide polymorphism) que ocorrem, em média, a cada 1000 bases, são as variações mais comuns que ocorrem no genoma. Como regra geral, os SNPs não têm efeito na função celular, entretanto, muitos deles estão altamente relacionados a doenças e a influência de fármacos nas respostas celular. A análise desses polimorfismos é de grande importância para vários campos relacionados à saúde, diagnóstico e comportamento fisiológico. Os polimorfismos de nucleotídeos únicos podem trazer importantes informações para a medicina esportiva e os estudos da aptidão física. De acordo com a literatura, diversos polimorfismos de genes relacionados a força muscular, sistema cardiovascular e ao metabolismo celular afetam a expressão ou a eficiência das proteínas codificadas por esses genes, promovendo maior ou menor aptidão ao exercício. Dessa forma o estudo de polimorfismos de genes envolvidos na performance atlética torna-se de grande importância para a caracterização de perfis genéticos ligados aos fenótipos de melhor desempenho para exercício físico, abrindo ainda a possibilidade de desenvolvimento de treinamentos específicos para alcance de desempenho máximo. Entretanto, para se avaliar a presença desses polimorfismos em nossa população, bastante miscigenada, temos que considerar também a frequência de determinado polimorfismo em relação ao seu grupo étnico. Desta forma, esta parte do projeto Atletas do Futuro tem como finalidade desenvolver um banco de dados genéticos de nossos atletas de elite, com o objetivo futuro de criar uma ferramenta como indicativo de desempenho físico que deverá ser utilizada na orientação de crianças em estágio inicial de atividade esportiva, orientação de tipo e carga de treinamento para atletas de elite e atletas de lazer.
