  Estudo eletrofisiológico e nociceptivo da toxina Ts8 proveniente da peçonha do escorpião Tityus serrulatus, Projeto certificado pelo(a) coordenador(a) Eliane Candiani Arantes em 16/11/2016., Descrição: Este projeto buscou estudar funcionalmente a toxina Ts8 utilizando ensaios eletrofisiológicos e de nocicepção. Ts8 demonstrou , entre 20 canais testados, ser uma toxina seletiva para canais para potássio Kv4.2. Com base em estudos que investigaram o envolvimento de Kv4.2 no controle da nocicepção, o efeito da toxina também foi avaliado na nocicepção. A administração intraplantar da toxina em animais demonstrou aumentar o comportamento nociceptivo (tempo de lambida e levantamento da pata), além de também induzir hiperalgesia mecânica. Ainda, a administração intratecal prolongou a hiperalgesia mecânica.   Purifcação e caracterização funcional de peptídeos lineares da peçonha de Tityus serrulatus, Projeto certificado pelo(a) coordenador(a) Eliane Candiani Arantes em 16/11/2016., Descrição: O presente projeto de pesquisa buscou purificar dois peptídeos lineares (sem pontes de dissulfeto) da peçonha do escorpião T. serrulatus (NDBPs, do inglês non-disulfide-bridged peptides), denominados venom peptídeos 7.2 (RLRSKG) e 8 (KIWRS). Adicionalmente, para facilitar os ensaios funcionais, esses peptídeos foram sintetizados, comparando com o peptídeo sintético já descrito 7.1 (RLRSKGKK). Os peptídeos sintéticos foram submetidos a uma variedade de ensaios biológicos: actividade hemolítica, antifúngica, antiviral, eletrofisiológica, imunológica e de inibição da enzima de conversão da angiotensina (ECA).   Caracterização estrutural e eletrofisiológica da toxina Ts4 da peçonha do escorpião Tityus serrulatus, Projeto certificado pelo(a) coordenador(a) Eliane Candiani Arantes em 16/11/2016., Descrição: Esse projeto de pesquisa buscou estudar a toxina Ts4, anteriormente descrita como uma toxina "não tóxica", presente na peçonha do escorpião Tityus serrulatus (Ts). A modelagem da toxina Ts4 foi realizada utilizando a estrutura conhecida da Ts1, a toxina majoritária da peçonha que apresenta sequência de amino ácidos com alta identidade com a Ts4 (65,57%). A caracterização eletrofisiológica foi realizada utilizando seis canais para sódio (Navs) de mamíferos (Nav1.2→Nav1.6 e Nav1.8) e dois canais para sódio de insetos (DmNav1 e BgNav). Os ensaios eletrofisiológicos mostraram que a toxina Ts4 inibiu especificamente a inativação rápida de canais Nav1.6, canal abundante no sistema nervoso central do adulto.   Caracterização eletrofisiológica e imunológica da toxina Ts5 da peçonha do escorpião Tityus serrulatus, Projeto certificado pelo(a) coordenador(a) Eliane Candiani Arantes em 16/11/2016., Descrição: A peçonha de Tityus serrulatus (Ts) é constituída principalmente por neurotoxinas específicas para canais para potássio (Kvs) e canais para sódio (Navs) voltagem-dependentes, os quais estão expressos em muitas células incluindo macrófagos. Os macrófagos são a primeira linha de defesa da imunidade inata e participam da resposta inflamatória no envenenamento por Ts. No entanto, pouco se sabe sobre o efeito das toxinas de Ts sobre a ativação de macrófagos. Esse projeto de pesquisa buscou investigar o efeito da toxina Ts5 em diferentes canais para sódio, bem como seu papel na imunomodulação de macrófagos. Os ensaios electrofisiológicos demonstraram que a toxina Ts5 inibe a rápida inactivação dos canais Nav1.2, Nav1.3, Nav1.4, Nav1.5, Nav1.6 e Nav1.7. Curiosamente, Ts5 também inibe a inativação do canal para sódio de Drosophila melanogaster do inseto (DmNav1), e é portanto classificado como a primeira toxina a-like. Os experimentos imunológicos em macrófagos revelaram que a toxina Ts5 é proinflamatória, induzindo a produção de TNF-alpha e IL-6. A nova toxina, denominada Ts19 Frag-II apresenta 49 resíduos de aminoácidos, três pontes dissulfureto, Mr5534 Da e foi classificada como um novo membro das beta-KTxs, a segunda descrita para o escorpiãoTs .
