Investigação dos efeitos e mecanismos de ação dos extratos de plantas nativas do NE Brasileiro, em modelos mamíferos visando o desenvolvimento de novos fármacos para tratamento de miopatias cardíacas e musculares alcoólicas e outras formas de intoxicação, Descrição: Preedy e colaboradores calcularam que miopatia alcoólica é a mais prevalente forma de doença muscular no mundo ocorrendo em mais de 50% daqueles que abusam do álcool mesmo que moderadamente. O etanol e seu principal subproduto, o acetil aldeído são inibidores potentes da síntese protéica muscular, e tanto proteínas contráteis como não contráteis são danificadas pelo álcool. Estes efeitos são causados por uso agudo e crônico que também causam redução da atividade do RNA muscular por mecanismos que envolvem o aumento das atividades da RNAse. Pesquisas recentes no Brasil indicam que o problema não é diferente no nosso país, e que de fato esse está se tornando um problema seríssimo para a saúde e economia . É possível que o extrato de pinhão-manso interaja com proteínas relacionadas ao processo agudo de reparo muscular, como por exemplo a proteína MG53 , e acelere o processo agudo de reparo (patching)? Store-operated calcium entry in muscle health, aging and fatigue, Descrição: During aging skeletal muscles become weaker and hundreds of millions of individuals become afflicted by a condition named sarcopenia. While some of the changes that take place in aged skeletal muscles are directly related to muscle atrophy, changes in contractile apparatus function cannot fully explain the decrease in the specific force during aging. Our recent studies revealed that muscle aging is associated with compromised store-operated calcium entry (SOCE, see figure below), the elemental process for refilling of the intracellular calcium stores in skeletal muscles and the majority of cells studied to date. As calcium (Ca) entry is chronically reduced in aged muscle fibers, Ca storage in the muscles decrease making less Ca available for each contraction-relaxation cycle, leading to reduced contractile force and fatigue. Thus, my lab was among the first to demonstrate that effective SOCE is essential for normal muscle contractility. Our research involves the use of genetically modified mouse models, molecular-genetic manipulations, muscle contractility studies, calcium imaging and a number of biochemical and biophysical approaches. We always combine in vitro with in vivo approaches to create a broader understanding of physiological phenomena.
