  Blue Muffin, Descrição: The present proposal builds on the Unpuzzle+ project, which was supported by the Lantmännen Research Foundation (20232024) and the Swedish Research Council (20222026). In the Unpuzzle+ project, we developed an analytical workflow to generate, purify and quantify oligomers (i.e., small pieces) of dietary fibre (DF) produced through enzymatic hydrolysis or during the gut microbiota fermentation process. Particularly, we focused on producing and isolating oligomers from wheat arabinoxylan (WAX) and oat beta glucan (OBG), and further confirmed their biological potential by exploring their interaction with receptors present in intestinal and immune system cells. The outcome of that project was a method that allows us to generate, isolate, and quantify these oligomers, whether they're produced by enzymes or by gut bacteria during fermentation. We also revealed how different oligomers can induce structure-dependent effects, and how their profile can change throughout gut microbiota fermentation and between individuals, using simulation of human colonic fermentation. Now, in the Blue Muffin study, we are expanding on this work to understand formation dynamics of bioactive oligomers in humans. We will define how factors like gut transit time, luminal environment and gut microbiota profile and functioning impact the production of these bioactive oligomers after ingestion of a DF-source food. This knowledge will allow us to determine key parameters to predict the formation of bioactive oligomers in humans that, combined with the knowledge on structural characterization and production of oligomers acquired in Unpuzzle+, will allow us to design DF and bioactive oligomers with desired health effects to specific individuals.   CoSMOSMiTE: Comprehensive screening of metabolites in organisms on space missions for technological exploration, Descrição: Exploring life beyond Earth raises questions about food production and lifespan changes in organisms metabolism. Plants are vital for human life providing food and oxygen. Moreover, understanding lifespan adaptation of human metabolism to extraterrestrial conditions is critical. Model organisms such as Arabidopsis and the worm Caenorhabditis elegans can be used as tools to understand physiological and metabolic process of plants and humans exposed to extraterrestrial conditions, respectively. In CoSMOSMiTE, we will apply metabolomics to investigate Arabidopsis and C. elegans aboard the International Space Station (ISS), shedding light on how to produce food and understanding dynamics of sustaining life beyond Earth.   MeET: Development of toolbox for exposome and metabolomics analyses, Descrição: The primary aim of the proposed project is to develop an analytical and computational pipeline for comprehensive characterization of chemical compounds, including both metabolites as well and exogeneous chemicals. Such a pipeline would be useful to inform public and environmental health authorities about the chemical safety, as well as provide a powerful platform for discovering novel disease biomarkers that may be amenable for the development of novel diagnostic applications. Overall, the project will create a platform for innovative, high-impact metabolomics and exposome research.   Unpuzzle: Unravelling molecular motifs from dietary fibers responsible for interaction with recognition receptors in human cells, Descrição: Dietary fibres (DF) can induce beneficial health effects by interacting with receptors at the surface of intestinal epithelial cells and immune system cells. Although the knowledge of the structure-function relationship between DF and their biological effects in human cells is highly desirable for successful translational applications, the molecular motifs within the often-complex DF structure responsible for binding to human cell receptors remain poorly understood. In Unpuzzle, we propose a new methodological framework to explore the interaction between DF and Toll-like receptors (TLR) in human cells. We will reduce the structural complexity of selected DF by generating oligomers through non-enzymatic hydrolysis. The produced oligomers (structural motifs) will be further fractionated and evaluated for interaction with TLR using reporter cells. The structural motifs that can interact with TLR will be characterized through high-resolution tandem mass spectrometry techniques and explored for biological effects in human macrophages through metabolomics-based approaches and cytokine array. Finally, network analysis will link structural data of molecular motifs with effects in human cells. Results will clarify how DF interacts with human cells and the potential consequences of such interaction on our gut health.   ImprUV(+): An analytical framework to explore the effects of UV during plant growth on human health, Descrição: The From Farm to Fork (FFF) strategy was designed to build a fair and sustainable food system. While FFF ticks many boxes in the sustainability agenda, it does not address how to improve crop nutritional quality. In ImprUV+, we are employing an analytical toolbox that goes beyond the FFF concept to explore whether ultraviolet light (UV) during plant growth improve the nutritional quality of horticultural crops. Firstly, we are applying targeted, semi-targeted and non-targeted metabolomics approaches to explore UV effects on plant metabolism but since plant health benefits are also dependent on human gut microbiota fermentation, we are also employing metabolomics to explore the extracts of control and UV-exposed plants obtained after fermentation by the human gut microbiota. Finally, reporter cell assays are applied to validate whether the fermentation products of control and UV-exposed plants differentially activate human intestinal receptors. By combining plant science, metabolomics and gut health, ImprUV+ will deepen our knowledge about plant responses to UV, while bringing a simple and energy-efficient strategy to improve crop nutritional quality and providing insights into potential benefits on human health.   ImprUV: Applying non-target metabolomics and network analysis to increase the sensory quality of vegetables and herbs, Descrição: What if science could make vegetables and herbs that are nutritious and taste good even better?ImprUV, a project from Örebro Universitys focus area Food and Health, explores how we, by using UV light, can improve the flavor and nutritional quality of horticultural products in an efficient, commercially sound, and sustainable way. To explore this, we propose the cutting-edge idea of combine plant, sensory, and medical sciences with metabolomics (i.e., the comprehensive analysis of compounds present in a biological system). By applying network analysis to link analytical data on plant metabolism with sensory analysis data and biological data, we aim to define how UV light modulates during plant growth specific metabolites related to the sensorial and nutritional quality of horticultural products.   Immunomodulatory potential of non-starch polysaccharides isolated from edible fruits and mushrooms, Descrição: Non-starch polysaccharides (NSPs) derived from certain plants and edible fungi can modulate immune function, contributing to health maintenance. The monosaccharide composition of NSPs, along with the phylogenetic proximity between organisms, suggests the occurrence of polysaccharides with immunomodulatory effects in other, yet unstudied, species. This hypothesis extends to the NSPs of banana, passion fruit, and the Pleurotus albidus mushroom, which may also exhibit these properties. The aim is to extract, purify, and characterize NSPs from these foods, test them in macrophage cultures, and assess their immunomodulatory action by measuring interleukin levels, nitric oxide (NO) production, and conducting phagocytosis assays. Additionally, NSPs from banana will be obtained at a pilot scale, considering the potential application as a functional ingredient. The results will enhance our understanding of the composition of these foods and aid in identifying fractions with potential use as food or pharmaceutical ingredients.
