Robert Frans Huibert Dekker

Graduated in 1969 with B.Sc. (Honours-biochemistry) from University of Western Australia, M.Sc. (1972) and Ph.D. (1975) in microbial biochemistry from James Cook University (AUSTRALIA), and post-doc (1975/76) at Freiburg University (GERMANY). Dr Dekker has worked in Australia, Germany, South Africa, Brazil, Spain and Canada over a 45-year period on biorefining and biotechnology-related research projects using biochemical (enzymes) and microbiological (fermentation) approaches. His research has focused on Applied Biochemistry and Microbiology, and Industrial Biotechnology. Specifically, his research themes have centered on: ruminant digestion of plant polysaccharides; polysaccharide bio/chemistry; enzymology; biosynthesis of bacterial cell wall/membrane components; fermentation technology; microbial physiology; enzyme applications to wine oenology; steam-explosion pretreatment of lignocellulosic materials; enzymatic saccharification of pretreated lignocellulosic residues; yeast fermentation of xylose into bioethanol and bioxylitol; enzyme immobilization onto magnetic supports and application in saccharification of steam-exploded cellulosic materials; screening of bacteria, yeasts and fungi for enzymes with applications in bioconversion of plant biomass; bioremediation of soils contaminated with polyaromatic hydrocarbons; lipase-catalyzed transesterification of plant seed oils to biodiesel; valorization of poplar bark (phytochemical extraction), value-addition of hemicelluloses; biological modification of Kraft lignin for polymer applications; and biological activity of fungal -glucans in animal models and mammalian cell-lines. In CANADA, Dr Dekker?s research was supported by competitive grants ($ 1.70 million) from Natural Science and Engineering Research Council (NSERC) of Canada, and Ontario Center for Research & Innovation in the Bioeconomy (CRIBE) in partnership with Canadian industry. During his career, he has been awarded competitive, peer-reviewed grant funding totalling $ 2,650,000.

    ? Valorização de Lignina: Degradação Enzimática de Lignina em Metabólitos Aromáticos de Baixo Peso Molecular, e Seleção de Bactérias com Atividade Ligninolítica, Descrição: A lignina constitui o biopolímero vegetal mais abundante na Terra depois da celulose. Apenas 2% da lignina disponível é comercializada, pois grande parte é utilizada na geração de energia de baixo custo, nas indústrias de papel e celulose. Este projeto objetiva valorizar a lignina, através de sua fragmentação por enzimas ligninolíticas fúngicas e bacterianas, para produzir compostos químicos úteis para indústrias químicas, cosméticas e farmacêuticas, sem agredir o ambiente. Um fungo ligninolítico previamente selecionado será cultivado em meio líquido contendo lignina, sob condições estática e submersa, para produzir as referidas enzimas. Os metabólitos aromáticos monoméricos e oligoméricos serão identificados por CLAE e as suas estruturas químicas determinadas por UV/Vis, FT-IR, LC-MS e RMN. As enzimas constituem uma excelente estratégia em processos de degradação porque diminuem etapas químicas, o uso de solventes e reagentes orgânicos, além de não poluírem o ambiente. Enzimas microbianas ainda não descritas poderão também ser descobertas, dado o pouco conhecimento sobre a degradação enzimática de ligações químicas da lignina. Bactérias de diversos ambientes serão isoladas, selecionadas e identificadas. Aquelas que forem capazes de crescer e degradar lignina em seus fragmentos e compostos aromáticos, serão avaliadas. A formação de novos pesquisadores na área de biorrefinamento será outra contribuição. This research proposal addresses the valorization of lignin through its fragmentation by fungal ligninolytic enzymes, and to screen for bacteria that can metabolize lignin. A ligninolytic fungus is grown on liquid nutrient media containing lignin under conditions of static and shaken cultivation, to produce enzymes that can fragment lignin into monomeric and oligomeric aromatic compounds. The products are identified by HPLC, and their chemical structures determined by spectroscopic methods (UV/Vis, FT-IR, LC-MS, NMR). Process technology will be developed to produce these chemicals by environmentally friendly methods. The expected outcomes are to produce new oligomeric compounds from lignin that will lead to new applications. Head of Biomass Program: Bioconversion of lignocellulosic residues. Researched the following areas: steam-explosion pretreatment of lignocellulosic residues (bagasse, sunflower seed hulls, softwood and hardwood residues); enzymatic saccharification of pretreated residues; pentose sugar fermentation to ethanol; immobilization of enzymes (cellobiases) onto magnetic supports and their application in the enzymatic saccharification of pretreated lignocellulosic residues. Developed a demostrated technology for the bioconversion of lignocellulosic residues into fermentable sugars in high yields using steam explosion pretreatment and cellulase saccharification. The method has potential for industrial development. During my time at CSIRO, I was invited as a Specialist Consultant to the United Nations (UNDP) project on bioconversion of lignocellulose in Chile, 1989.
