Matias Eliseo Melendez

Bolsista Produtividade em Pesquisa CNPq (Nível PQ-2). Possui graduação em Licenciatura en Genética - Universidad Nacional Misiones (Argentina - 2004) e mestrado em Genética e Biologia Molecular pela Universidade Federal do Rio Grande do Sul (Brasil - 2006). Apos um período de um ano trabalhando com a proteína pro-apoptótica TRAIL/Apo2L na National University of Ireland - Galway (NCBES-NUI Galway-Irlanda - 2007), realizou o seu doutorado na Université Claude Bernard Lyon 1 (França - 2007_2011) trabalhando na produção de vetores virais derivados do vírus da Herpes Simplex Humana de tipo 1. Posteriormente realizou pós-doutorado (FAPESP) no Hospital de Câncer de Barretos (Brasil), trabalhando na área de terapia gênica oncolítica. Atualmente é Professor e Pesquisador no Instituto Nacional de Câncer (INCA), no Rio de Janeiro, Brasil. Tem experiência na área de Genética e Biologia Molecular e Celular. Atualmente trabalha no desenvolvimento de vetores virais derivados do virus HSV-1, desenvolvimento de proteinas recombinantes derivadas de glândulas veneníferas e inovação em diagnóstico molecular.

PRODUÇÃO, PURIFICAÇÃO E ANÁLISE IN VITRO DE UMA NOVA ISOFORMA RECOMBINANTE SOLÚVEL DO INDUTOR DE MORTE CELULAR APO2L/TRAIL, Descrição: O emprego intensivo dos protocolos pré e pós-cirúrgicos para tratamento do câncer vem melhorando os resultados do tratamento de alguns tipos tumorais. Entretanto, muitos tumores ainda apresentam uma baixa resposta aos protocolos clínicos, limitando a indicação e a eficácia do tratamento. Por este motivo, se fazem necessárias novas modalidades terapêuticas mais eficientes, que possam oferecer aos pacientes oportunidades reais de controle de fácil administração e com menores efeitos colaterais. Neste sentido, o uso de indutores protéicos de morte celular por apoptose vem sendo aplicado em ensaios clínicos, dado que possuem grande potencial terapêutico. Assim, o nosso laboratório construiu e desenvolveu uma nova isoforma solúvel do indutor de morte celular Apo2L/TRAIL, nomeada sfTRAIL. A proteína recombinante sfTRAIL possui um domínio de trimerização nunca antes descrito para esta proteína, que a torna ativa em suspenção, sendo capaz de promover apoptose em linhagens celulares sensíveis a Apo2L/TRAIL. Esta proteína possui grande potencial inovador e terapêutico para tratamento de tumores sólidos, a qual poderia controlar e eliminar as células neoplásicas, retardando ou impedindo a recorrência tumoral. Os resultados prévios do nosso grupo com esta nova proteína potencialmente terapêutica justificam os principais objetivos do trabalho: (i) construir uma linhagem celular que expresse de maneira estável a proteína sfTRAIL do sobrenadante, (ii) purificar por cromatografia de afinidade da proteína recombinante sfTRAIL, (iii) aprofundar a caracterização da proteína recombinate sfTRAIL e (iv) analisar in vitro a eficiência da proteína recombinante purificada. Plataforma brasileira para estudos de bioprospecção de compostos naturais com potencial anticâncer em fase pré-clínica, Descrição: O câncer é responsável por um número significativo e crescente de pacientes no mundo todo, e representa a segunda causa de morte da população mundial. As neoplasias em mama, pulmão e colo uterino representam tumores sólidos mais frequentes em adultos cujos tratamentos não sofreram grandes avanços, resultando em altos índices de mortalidade dentre os pacientes. Dessa forma, a busca de terapias alternativas e complementares tem sido uma constante. A tentativa de aprimorar a terapia antitumoral com novos agentes antineoplásicos de origem natural tem revelado eficácia e oferece um grande campo para investigação científica. As estratégias para a descoberta de novos fármacos têm mudado ao longo dos anos. Os programas de prospecção incluem testes in vitro em linhagens tumorais humanas o que tem resultado em um elevado número de moléculas promissoras. Um programa de bioprospecção que contempla avaliação da citotoxicidade em linhagens tumorais humanas utilizando um novo composto semi-sintético derivado da plantas está sendo desenvolvido em nosso laboratório. Até o momento, atividades biológicas incluindo, mais recentemente, potencial antiproliferativo em câncer gástrico foram descritos para os fitocompostos. Inserido neste contexto, propomos aqui o estabelecimento uma plataforma brasileira que permita avaliar e caracterizar de forma abrangente estudos de bioprospecção de compostos naturais com potencial anticâncer em fase pré-clínica. Este estudo propõe a obtenção de modelos in vitro e in vivo, utilizando uma plataforma de maior representatividade nos tipos tumorais de mama, pulmão e colo uterino, compreendendo linhagens estabelecidas e primárias. Propomos ainda adequar essas culturas ao modelo 3D como método de varredura de alto desempenho capaz de mimetizar de forma mais eficiente o microambiente tumoral. Em paralelo, objetivamos caracterizar molecularmente a heterogeneidade dessas linhagens a partir de informação genômica com dados de Next Generation Sequencing (NGS) e expressão gênica em larga escala usando a plataforma Nanostring. Além do mais, objetivamos a caracterização funcional e a validação dos principais alvos por meio de uma biblioteca de siRNA.
